1 .Indications

1. Urinary system diseases, such as acute and chronic glomerulonephritis, acute and chronic pyelonephritis, acute and chronic renal failure,

nephrotic syndrome, cystitis, urethritis, urinary calculi, urinary tumors, etc.

2. Systemic diseases with secondary renal damage, such as diabetic nephropathy, lupus nephropathy, hypertension and arteriosclerosis renal damage,

multiple myeloma renal damage, etc.

3. Diseases of other systems or organs, such as diabetes, hemolytic anemia, jaundice hepatitis, water and electrolyte or acid-base balance disorders, etc.

4. Treatment monitoring of nephrotoxic drugs: such as aminoglycoside antibiotics (such as gentamicin, kanamycin, tobramycin), some third-generation

cephalosporins, sulfonamides, some immunosuppressants .

5. Health checkup, such as occupational disease monitoring for kidney damage caused by exposure to heavy metals.

2. Specimen collection

1. Preparation before collection: The patient drinks water normally, and the user should indicate the type and time of the drug. Before collecting urine,

clean the collection site, including the urethra and surrounding skin, to avoid the influence of menstruation, vaginal secretions, prostatic fluid, semen,

smegma, feces, etc., and cannot be collected from the urinal.

2. Prepare a container for collecting urine: generally a special inert material product, such as a disposable transparent plastic cup, preferably with a lid,

and the volume of the cup should be greater than 50ml. If you want to check the urine volume of 12 or 24 hours, you should use a transparent glass of more than 500ml.

Glass bottles, but must be clean, dry, and leak-free.

3. Collect urine

⑴Morning urine: It is collected during the first urination after getting up in the morning, which is the first morning urine. The results may be affected by the fact

that urine stays in the bladder for too long (greater than 8 hours) and sugar is broken down. Pay attention to dust and pollution after collection, and submit them for inspection within 1 hour.

(2) Random urine: urine samples collected at any time, suitable for both outpatient and emergency patients, and can be used for the examination of urine chemical

components and tangible components, but can only reflect changes in urine at the time of collection, and is susceptible to urine samples. A variety of factors, such as diet,

water intake, exercise status, etc., some pathological changes may not be easily detected, leading to missed diagnosis, and the results of each random urine test may vary greatly,

with poor repeatability, and sometimes even cause clinical symptoms. The results are confusing.

(3) Urine three-cup test specimens: One urine specimen is collected in three times, namely the initial urine, the middle urine and the final urine, which are respectively placed in three

transparent plastic urine specimen collection cups for inspection.

4. Preservation of urine

⑴Preservation: Due to some reasons, the specimen cannot be checked in time after collection, and chemical preservatives can be added for preservation. 5~10ml of 40% formaldehyde

can be added to 1000ml of urine, which is suitable for the preservation of formed components of urine. Add 5~20ml of toluene to 1000ml of urine to isolate the air on the liquid surface,

which can be used for the determination of urine sugar or urine protein.

⑵ preservation time: 2 ~ 8 ℃ generally not more than 6h, the time is too long can still cause damage to the components in the urine. When refrigerated, urate, phosphate, etc. are easy

to crystallize out, which can interfere with microscopic examination of formed components.
